Orlistat is a long-acting and potent specific gastrointestinal lipase inhibitor. It is white or white-like powder at room temperature, insoluble in water, soluble in chloroform and soluble in ethanol. It inactivates the enzyme by forming covalent bonds with the active serine sites of gastric lipase and pancreatic lipase in the stomach and small intestine. Fat in food cannot be broken down into free fatty acids and monoacylglycerol, so fat cannot be absorbed and utilized, thus reducing the body's calorie intake and controlling body weight. The drug works without absorption throughout the body. Fat absorption can be inhibited by 30% at the usual dose. Rarely absorbed after oral administration, it can be metabolically inactivated in the intestinal tract, the metabolic site is in the gastrointestinal wall, and the elimination half-life is about 14 to 19 hours. About 97% of this product is excreted in feces, of which 83% is excreted in its original form. Clinically orlistat can be used in obesity and hyperlipidemia.
